Unit Description

Laser lines

The LSM 710 is capable of 34 spectral detection channels using six laser lines - 405, 458, 488, 514, 561 & 633 nm, plus an additional 594 nm HeNe laser (includes replacement main beamsplitter wheel equipped for 458, 488, 514, 561, 594 & 633 nm laser lines). The 561 and 594 lasers are needed to visualize mCherry, Texas Red, and other red fluorophores including Alexa 568, Alexa 594.

Fluorescence Equipment

The LSM 710 uses a six position turret that houses cubes holding a DAPI fluorescence filter, a Cy3 fluorescence filter and a GFP fluorescence filter.

Transmitted light equipment

The transmitted light source uses a halogen bulb for transmitted light source and a mirror to switch between transmitted light source and transmitted PMT (detector).

HBO Illuminator

The fluorescence light source uses a mercury bulb and power supply along with a touch pad for microscope control with docking station.

Manual Stage

The stage for microscope has specimen holders for slides and petri dishes allowing for imaging of fixed and live cell samples.

Lens Objectives

The LSM 710 has 10X, 20X, 40X, 63X water and 100X oil objectives and Nomarski optics.

Environmental Control System

An incubator chamber encloses the microscope allowing for temperature control during image acquisition. A heated insert for the stage holds petri dishes and a cover for this insert delivers CO2. The Environmental Control Chamber can be used for containment of mammalian cells and C.elegans allowing for motility studies. For temperature sensitive experiments the microscope stage can be cooled allowing for a working temperature range of 4°C to 45°C.

Computer driven X-Y stage

The computer driven X-Y stage allows imaging of multiple fields over time for any given live specimen. It is ideal for projects that collect multiple images over a period of time intervals or require imaging of multiple animals (C. elegans, Drosophila, mammalian cells). This stage allows for imaging to take place in parallel, rather than serially, facilitating collection of more data over the length of the experiment.

FRET/FRAP software package

The LSM 710 is fitted with quantitative FRET/FRAP (Fluorescence Resonance Energy Transfer/ Fluorescence Recovery after Photobleaching) software package allowing for acquisition and analysis of sensitized emission, acceptor photobleaching and Emission fingerprinting based FRET and FRAP fitting parameters to establish rate constants for recovery.

Image VisArt plus program module

Permits innovative 3D and 4D renderings of the data. Up to three clipping planes allow different rendering modes within the same sample.

Typhoon FLA 9500 is a versatile laser scanner that is ideal for imaging of fluorescent Western Blot membranes. Biomolecular imaging applications include sensitive and quantitative measurements of Western blots, multiplex fluorescence and radioactive labels. It can detect more than one protein at the same time by means of secondary antibodies labeled with different fluorophores. Amersham ECL Plex fluorescence detection systems are compatible with typhoon FLA 9500. It contains the 2-D DIGE system for accurate quantitation of changes in protein expression and the DeCyder 2D differential analysis software. High sensitivity and broad dynamic range make it highly suitable for 2-D DIGE applications to detect and quantitate subtle changes in protein expression.

Typhoon FLA 9500 features

  • Imaging of multifluorescent, chemifluorescent, radioisotope-labeled and calorimetric samples
  • Provides 457/532/635 nm excitation and up to 10 ?m resolution for gel, blot, tissue and array imaging. The system is customized with 785 nm excitation, filters and detectors for multiplex and near infrared fluorescence applications.
  • High resolution and quantitation: a pixel resolution of 10 ?m and a linear signal response over five orders of magnitude provides precise quantitation in gels, blots, tissue sections and arrays
  • High sample throughput: scanning area of 40 x 46 cm enables simultaneous imaging of up to 20 gels or blots measuring 10 x 8 cm in size.
  • 2-D DIGE imaging: simultaneous imaging of two 2-D DIGE gels for differential expression studies
  • Visible and infrared fluorescence imaging: near infrared excitation for imaging IRDye and other infrared dyes.
