Abstract:
A major driver of type 2 diabetes hyperglycemia is increased glucose production from non-carbohydrate precursors via hepatic gluconeogenesis. While many substrates can serve as carbon sources for gluconeogenesis, their exact quantitative contributions to the process remain less defined. Here, we will discuss an underappreciated role of circulating glycerol as a mediator of increased hepatic glucose production in type 2 diabetes and a potential therapeutic target. Data will be presented from preclinical and clinical experiments using stable isotope tracer infusions and mass spectrometry analysis.
